Most vaccination audits inspect a folder. They confirm that the right product left the right fridge on the right day, which is worth knowing and proves almost nothing. The question that decides whether the programme worked is whether the dose reached the bird, and it is answered somewhere else entirely.

Layer one: what the paperwork can and cannot prove
The document layer is the easiest to build and the easiest to over-trust. What belongs in it is well established: vaccine batch numbers recorded against the flock or the hatch, storage temperature history, diluent identity and volume, mixing times, expiry dates, and the identity of the person who prepared the dose. Hatchery guidance is explicit that the batch number is recorded for traceability at the moment of use, not reconstructed afterwards.
Two preparation details deserve their own line in the checklist because they invalidate everything downstream when missed. Water sanitisers are stopped ahead of a drinking water vaccination, with common guidance putting that at 24 hours, since residual disinfectant destroys a live vaccine in the line. And where a stabiliser is used, skimmed milk powder at roughly 2 to 2.5 grams per litre is mixed into the water some 20 minutes before the vaccine goes in, not at the same moment.
All of that is necessary. None of it is evidence of vaccination. A perfectly documented dose can still end up in the albumen instead of the amnion, on the floor of a chick box instead of on the chicks, or in a drinker line that half the flock never reached.
Layer two: watching the application while it happens
This is where audits earn their cost, and it requires someone standing at the machine with a clipboard rather than reading a report afterwards.
In ovo: deposition site is the whole question
Injecting the egg is not the same as injecting the embryo. A field trial presented at the 2009 Poultry Science Association meeting, and designed with the poultry technical services team of the company marketing one of the systems, compared deposition accuracy between two commercial in ovo devices: 830 of 871 viable eggs, or 95.3%, were injected at the proper site with the automated system, against 486 eggs, or 52.3%, with the manual one. Read that as a demonstration of how wide the spread between application methods can be, rather than as an independent purchasing benchmark, since one of the two systems belonged to the sponsor.
The audit tool that costs nothing here is the residue breakout after hatch. Candling and breakout analysis identify eggs that were set upside down, whose embryos are effectively killed by the needle during in ovo injection, and that count is a direct measure of how well the setting stage fed the injector. Spray: coverage is measurable in thirty seconds
Hatchery guidance describes a check any supervisor can run: place a sheet of absorbent paper inside an empty chick box, pass it under the cabinet, and look at the pattern. It shows whether the whole surface of the box is covered evenly and whether vaccine is being wasted outside it. Alongside it sit the nozzle and pressure settings, which must produce a coarse spray of droplets at least 100 to 150 microns across, and the water itself, which is expected to be unchlorinated, low in minerals and taken from a container that has never held disinfectant.
Drinking water: the line is the equipment
On this route the audit follows the water rather than the machine. The vaccinated volume is calculated to be drunk within roughly one to two hours, so the observation is simply whether the line empties in that window and whether birds at the far end drank at all. Header tanks, long runs and dead legs are where coverage quietly disappears.
Layer three: the flock answers several weeks later
Serology is the only layer that reports on the birds themselves, and it reports late. For an inactivated vaccine, veterinary guidance places the sampling no earlier than four weeks after administration, because an earlier bleed reads an incomplete response and invites the wrong conclusion.
What matters in the result is less the average titre than its spread across the sampled birds, expressed as the coefficient of variation. Uniformity is what application quality produces, and unevenness is what a badly applied vaccine looks like in a laboratory report.

These bands are read by the laboratory and the veterinarian, against the specific test, the vaccine and the flock’s history. They are shown here so that a production manager can follow the discussion, not so that a hatchery or a farm can interpret a serology report on its own.
Making the audit repeatable
An audit is only useful in a series. That means fixing the sample points before the first visit, observing the same operations rather than whoever happens to be on shift, and keeping the questions identical from one round to the next so that the comparison survives a change of auditor.
It also means writing the corrective action next to the finding, with a date and a name. The most common failure of vaccination audits in this sector is not a missing check, it is a finding that nobody owned. When the same observation appears twice, the audit has stopped measuring the flock and started measuring the follow-up.
